Route Created but not reflected.

I have created a route and assigned a aircraft to that route but at Airline Overview, it did not reflect it as departing soon

Not at the moment, as an aircraft can only be scheduled on one route per 24 hour period, you could try SIN-BKK-SIN-SFO-SIN
However you may not get that once per day due to flight time and turnaround time required.

I actually found a strategy to avoid the half scheduling routes. You need separate your route into multiple routes.
 
Now when you activate a your half scheduling route, the game will only schedule two legs so make sure you only have two different flight segments per route so that they will schedule correctly when activated. For example XXX-XXX-XXX as one route and then XXX-XXX-XXX as another route and so on. In your case you should have two routes, POA-PVH-POA as one route and POA-EZE-POA as the other. In order for them to fly on the same day you should make the departure time for POA-EZE-POA at least ten minutes after the ready time for the end of POA-PVH-POA. If you want to be extra careful so to make sure that the first leg doesn't repeat itself, then you can activate the first leg and then immediately deactivate it, this way the first leg will schedule but only once until activated again.

I know this is tedious, but it's probably the best way to combat the half scheduling problem until Unknown98 can find a fix.

You need to make it all one route. Not separate routes.

POA-PVH-POA-EZE-POA

A route is like a daily schedule for an aircraft so in most cases you can only have 1 route assigned per aircraft.
